Histologic Changes after Intracavernous Injections of Vasoactive Agents, Solutions with Different Osmolarity and pH in the Rabbit




Abstract
We investigated histologic changes following intracavernous injection of vasoactive agents and solutions with different osmolarity and pH in rabbits.
Initially, four groups of three rabbits each received intracavernous injections of the following agents once a week for six consecutive weeks: group 1, normal saline 1ml, group 2, papaverine 20mg/ml, group 3, phentolamine 5mg/ml, group 4, PGE1
20§¶/ml.
One week after the last injection, the penises were histologically examined with H-E and Masson's trichrome stain. Next, another four groups of three rabbits each received the following solutions using the previous method: group 1, 320mosmol/l
and
pH 2,
group 2,320mosmonl/l and pH 7, group 3,3200 mosmol/l and pH 2 and group 4, 3200mosmol/l and pH 7.
Moderate to mild fibrosis was found after the injection of papaverine and mild to none after phentolamine and PGE1. Fibrosis was not noted after normal saline as well as solutions with different osmolarity and pH.
These findings suggest that penile fibrosis following intracavernous injection results from a property inherent in the injected drug and not from the pH or osmolarity of the solutions.
